My speaker has a + and - Terminal. HOw do I determine which is what from the Amp? Is there a DC voltage present on the wires from the amp that I could check or something else?

if there is a speaker cable already plugged into the amp, the sleeve is negative and the tip is positive. first, use a 1 1/2volt battery and with two wires, connect it to the speaker terminals. when the speaker moves forward, that is the proper connection.

if there is no speaker cable already plugged in, you may need to see the coloring of the wires on the output jack. the one connected to the ground connection would be the negative and the other the positive.

if they are reversed, it shouldn't hurt the amp but it will lose some of the highs and the lows will sound weird. turn off the amp, and reverse them if this happens.

the wires come directly out of the amp .. no plug. I know where the + and - are on the speaker , its labeled. the wires out of the amp are not labeled.

What kind of amp and are the wires colored? It really makes no difference on a single speaker as it only decides which way the speaker moves first.

David is quite correct. The polarity of speakers (plus and minus) makes absolutely no difference unless the amp has multiple speakers, or you're using two similar amps for the same instrument.

What David and Donny said. It makes NO difference how you hook up one speaker. Polarity only comes into play with multiple speakers.
