The low a screw on my c6 neck is too lose to the opening, it's really tough to get the tuning wrench into the nut. Is it ok to move it up into the hole above it? Also how hard is it to move? Do I just unscrew it and move it up?

Moving it up a hole should be fine. Assuming you're talking about the 10th string C-A change, that has quite a short throw. An even easier fix, if possible on your guitar, is to turn the guitar over and pull up on the middle of the rod enough to bend it until tip the tuning nut is easier to get to.
