I have read about a locking lever that some universal guitars have. Is this something that can be added to a standard E9 S-10 pedal steel, and if so does it matter if it is used on a pedal rather than a knee lever?

I'd just use a brick. Cool

Seriously, the mechanical implementation of a lock lever varies from one brand of guitar to another. There's no real "standard" way of doing it. Still, anyone skilled in tinkering with pedal steel machinery could add a lock to a pedal as easily as to a knee lever.

My guitar doesn't have any lock levers. Recently I was working on a recording that required holding in a lever for the entire solo. I just retuned the strings instead.
