Can someone tell my how to get this file? My shortcuts on my desktop doesn't work because of this missing file.

iexplore.exe is the main Internet Explorer exe (executable) program. If the link can't find that program it means it is either pointing to a wrong location for that file or the file has been deleted.

If you're using Internet Explorer and everything is working correctly, the link is wrong on that shortcut and you need to update it with the correct location. If you've updated to a different browser then the new browser wiped out (at least some) the Internet Explorer files. Updating Internet Explorer to a newer version should not cause this problem as I just updated to the new I.E. 5.5 and it still has the same links.

Do a find (click on start and then find and then files) and enter the iexplore.exe and see if it finds it on your computer. If it does, you can update your desktop shortcut to the correct location.
